“TransMPRA: A framework for assaying the role of many
trans-acting factors at many enhancers”

Gene regulation can be partitioned into contributions attributable to cis-acting elements (e.g., enhancers) and trans-acting factors (e.g., transcription factors). Genomic assays have revolutionized how we understand this cis-acting component of gene regulation. However, because of the sheer number of interaction tests, the lack of efficient perturbation methods to establish causality, and the complexity of gene regulation, high-throughput methods have largely ignored contributions from trans-acting factors. Here I will describe a new method for testing protein-enhancer regulatory interactions called transMPRA (trans Massively Parallel Reporter Assay) that combines the power of massively parallel reporter assays with perturbations of CRISPR screens. With transMPRA, I aim to characterize the trans-acting regulatory architecture underlying complex traits.
